| 体外研究 | Ferrostatin-1是HT-1080细胞中最有效的erastin诱导的铁蛋白沉积抑制剂（EC50 = 60 nM）。 Ferrostatin-1不抑制ERK磷酸化或阻止HT-1080细胞的增殖。然而，Ferrostatin-1可以防止erastin诱导的细胞溶质和脂质ROS的积累[1]。用0.4μMFerostostatin-1预处理的细胞显着降低细胞内活性氧（ROS）和氮物种（RNS）低于基础水平。另外，通过0.4μM的Ferrostatin-1预处理，细胞内ROS水平的增加也显着降低至基础水平以下。与载体（0.02％DMSO）处理的细胞相比，Ferrostatin-1处理24小时不会改变SHSY-5Y细胞中i-NOS的表达水平[2]。 |
| 激酶实验 | 用磷酸盐缓冲盐水（PBS）洗涤用Ferrostatin-1处理24小时的细胞三次，并通过离心沉淀。除去上清液，向细胞中加入80μL裂解缓冲液，然后在-20℃下储存过夜。随后，将细胞以10,000RPM离心12分钟，并将沉淀和上清液储存以备将来使用[2]。 |
